I have been taught three methods of setting levels, but I am unclear as to which one is best. One method is to move the RGB highlight and shadow sliders up to the first trace of data indicated in the hologram. The next method involves doing this to each color channel individually. The third method uses the eyedroppers to select the brightest and darkest neutral points within the image.

Which of these is best? Does it REALLY matter?

depends on the image. Doing each color channel is just like hitting auto levels.
i would just do the RGB channel.then use the grey dropper on somthing you know in the image is grey. then tweak any channel as necessary to fine tune color.
