What are some good general guidelines for when the sharpening tools should be used? After scanning, resizing, etc.?

If the image NEEDS sharpening, use the Unsharp Mask.
Do this in the final stage e.g. never resize after sharpening. When it is for print try a radius of 0.7 and when it is for screen (web), try a somewhat higher value.

Experiment with Threshold while keeping an eye on sufaces and noise.
