you can Save Selection, and call it back up later.
Matter of fact, if you save image as .psd or .tif, you can save saved selections with the image (alpha channel option) and call selections back up even after closing and reopening the image.

Alternatively, if you don’t make another selection, you can use Select|Reselect. It’s pretty neat in that you can do just about anything to the image after you have deselected, but as long as you haven’t made another selection, Reselect (Ctrl-Shift-D) will select your last selection.

Neat idea! It’s always been a pain to try and catch the same pixel when comparing adjustments at a particular place, especially when trying to set an RGB set of values.
Rule of thumb (I first heard read it as written by Bruce Fraser):
If a selection takes more than 30 seconds, save it as an Alpha channel.
I’ve created an action and assigned a key-combo to do just that with selections I want to save.
Oh, and the Command + Shift + D dealie works a treat, as long as you use it enough to remember that it’s available to you.

Forgive me if I am wrong (English is not my native language) but you are telling Cleveland how to save a selection. I was under the impression he asked how to save a selected pixel color (RGB values).
If my assumption is true then I would suggest
a) saving the color as a swatch or even better
b) using the color sampler tool
(Using the latter this also tells him visually where the pixel is located) Regards,
