Most file formats–JPEG, BMP, and so on–do not allow transparency. Period. If you save a transparent image in a file format that does not support transparency, it will not be transparent, and there is nothing you can do about it.

For example, if you take that Photoshop file and save it as a JPEG, it won’t be transparent and nothing you can do will make it transparent; the specification for the JPEG file format forbid transparency.

A GIF can be transparent; to save a transparent GIF, you use the "Save for Web" command.

However, it is then the responsibility of your program to display that transparency correctly.

What are you using the image for? Are you trying to make a transparent icon? If so, don’t use Photoshop! Transparency of an icon is created by using a mask. Download an icon creating program like IconFactory or icon Forge or AXXIS Icons,a nd use it to make your icon. There are plug-ins for Photoshop to create icons, but using Photoshop for creating icons is an exercise in frustration; a dedicated icon-creation program works much, much better.
