Make a layer clear not white

Can you assist? I need help with something which must be simple, I’ve almost got it but need a push over the edge. I’m editing a sequence with Premiere CS3 and need to paint an object out of the background. I import the clip into After Effects CS3, then import a still into Phtotshop CS3. Here I create a layer and ‘paint’ out a portion of the background. I then have in that layer my painted out area and a white screen. When I move this layer back to after effect I can motion track the layer no problems however I import not only the ‘painted area’ but also the white screen background, so I can only see what’s happening by turning down the layer’s opacity. I want to be able to import just the painted area of the layer not the white screen. It has to be simple but I can’t figure it please assist if you are able.

You need to ask this in the After Effects forum where they will no doubt know all about transparency and mattes in that application.
