What I am wondering is if I should save my photoshop file as a high quality jpg file then slice that, or should I just slice the Photoshop file?

I recommend you do all your work in Photoshop format. ImageReady will make the conversions to GIF (or JPG) when you are ready to publish it. Everytime you save a file in the JPG format, you can introduce noise and lose image quality. By keeping the original in PSD format, you will have a pristine version for later modifications.
